style=\"font-weight: 400;\">Saat puasa, tubuh beralih dari metabolisme glukosa ke biomolekul\/building blocks lainnya. Dalam 8\u201312 jam pertama, glikogen hati diurai menjadi glukosa melalui glikogenolisis (Berg et al., 2002). Setelah cadangan glikogen habis, glukoneogenesis (pembentukan glukosa baru dari asam amino dan gliserol) di hati menjadi sumber utama glukosa untuk otak (Owen et al., 1967). Namun, proses ini tidak efisien dalam jangka panjang. Tubuh mengaktifkan ketogenesis yang menghasilkan keton (\u03b2-hidroksibutirat dan asetoasetat) dari asam lemak di mitokondria hati (Cahill, 2006). Keton menjadi bahan bakar utama otak, jantung, dan otot, sehingga mengurangi ketergantungan pada glukosa.<\/span><\/p>\n<h3><b>Adaptasi Hormonal: Insulin, Glukagon, dan Growth Hormone<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Penurunan insulin dan peningkatan glukagon selama puasa mengaktifkan lipolisis (pemecahan trigliserida menjadi asam lemak) di jaringan adiposa (Boron &amp; Boulpaep, 2017). Asam lemak kemudian dioksidasi di hati dan otot untuk menghasilkan ATP. Selain itu, peningkatan growth hormone (GH) membantu mempertahankan massa otot dengan merangsang sintesis protein dan mobilisasi lemak (Ho et al., 1988). Kombinasi respons hormonal ini memastikan ketersediaan energi tanpa menghabiskan protein struktural.<\/span><\/p>\n<h3><b>Mitokondria: Optimasi Produksi ATP<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Puasa meningkatkan efisiensi mitokondria melalui <\/span><span style=\"font-weight: 400;\">mitochondrial biogenesis<\/span><span style=\"font-weight: 400;\"> (pembentukan mitokondria baru) yang dipicu oleh aktivasi <\/span><span style=\"font-weight: 400;\">PGC-1\u03b1<\/span><span style=\"font-weight: 400;\"> (peroxisome proliferator-activated receptor gamma coactivator 1-alpha) (L\u00f3pez-Lluch et al., 2006). Adaptasi ini meningkatkan kapasitas oksidatif sel dan mengurangi produksi <\/span><span style=\"font-weight: 400;\">reactive oxygen species<\/span><span style=\"font-weight: 400;\"> (ROS), menekan stres oksidatif.<\/span><\/p>\n<h3><b>Kesimpulan<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Puasa memicu serangkaian adaptasi seluler yang kompleks: peralihan metabolik ke keton, aktivasi autofagi, regulasi hormonal, dan peningkatan fungsi mitokondria. Mekanisme ini tidak hanya mempertahankan homeostasis energi tetapi juga berpotensi meningkatkan ketahanan sel terhadap stres patologis.
