400;\">Indonesia, sebagai salah satu <\/span><span style=\"font-weight: 400;\">megadiverse country<\/span><span style=\"font-weight: 400;\">, menyimpan 17% keanekaragaman hayati global (Mittermeier et al., 2011). Namun, ancaman deforestasi, perburuan liar, dan perubahan iklim mempercepat laju kepunahan spesies. Biologi konservasi memerlukan pendekatan inovatif untuk memantau dan melindungi keanekaragaman hayati secara efektif. Salah satu terobosan terkini adalah teknologi DNA Lingkungan (environmental DNA\/eDNA), yang menawarkan metode non-invasif untuk mendeteksi keberadaan spesies melalui sampel lingkungan seperti air, tanah, atau udara (Thomsen &amp; Willerslev, 2015). Artikel ini membahas peran eDNA dalam konservasi, aplikasinya di Indonesia, serta tantangan dan prospeknya.<\/span><\/p>\n<p><b>Apa Itu DNA Lingkungan?<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">eDNA merujuk pada materi genetik yang dilepaskan organisme ke lingkungan melalui kulit, lendir, feses, atau jaringan mati. Dengan teknik seperti <\/span><span style=\"font-weight: 400;\">metabarcoding<\/span><span style=\"font-weight: 400;\"> dan <\/span><span style=\"font-weight: 400;\">next-generation sequencing (NGS)<\/span><span style=\"font-weight: 400;\">, DNA ini dapat diidentifikasi hingga tingkat spesies, bahkan dalam konsentrasi rendah (Deiner et al., 2017). Metode ini mengatasi keterbatasan survei konvensional yang memakan waktu, mahal, dan kurang sensitif untuk spesies kriptik atau langka.<\/span><\/p>\n<p><b>Aplikasi eDNA dalam Konservasi Indonesia<\/b><\/p>\n<ol>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Pemantauan Spesies Prioritas<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">eDNA efektif mendeteksi satwa langka seperti Harimau Sumatera (<\/span><span style=\"font-weight: 400;\">Panthera tigris sumatrae<\/span><span style=\"font-weight: 400;\">) dan Badak Jawa (<\/span><span style=\"font-weight: 400;\">Rhinoceros sondaicus<\/span><span style=\"font-weight: 400;\">). Di Taman Nasional Ujung Kulon, analisis eDNA dari sumber air membantu memetakan distribusi Badak Jawa tanpa gangguan fisik.<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Deteksi Dini Spesies Asing Invasif<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">eDNA berperan kritis dalam mendeteksi introduksi spesies asing, seperti ikan Sapu-sapu (<\/span><span style=\"font-weight: 400;\">Pterygoplichthys spp.<\/span><span style=\"font-weight: 400;\">) di Sungai Ciliwung, sebelum populasi mereka meledak dan merusak ekosistem lokal (Wiryawan et al., 2021).<\/span><\/li>\n<\/ol>\n<p><b>Tantangan dan Keterbatasan<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Meski menjanjikan, implementasi eDNA di Indonesia menghadapi beberapa kendala:<\/span><\/p>\n<ul>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Kontaminasi Sampel<\/b><span style=\"font-weight: 400;\">: Sensitivitas tinggi eDNA rentan terhadap kontaminasi selama pengambilan sampel (Goldberg et al., 2016).<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Basis Data Referensi Terbatas<\/b><span style=\"font-weight: 400;\">: Database genetik lokal seperti <\/span><span style=\"font-weight: 400;\">BOLD Systems<\/span><span style=\"font-weight: 400;\"> masih minim, menyulitkan identifikasi spesies endemik (Andayani et al., 2021).<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Infrastruktur dan Biaya<\/b><span style=\"font-weight: 400;\">: Analisis NGS memerlukan fasilitas <\/span><span style=\"font-weight: 400;\">bioinformatika<\/span><span style=\"font-weight: 400;\"> yang belum merata di seluruh Indonesia.<\/span><\/li>\n<\/ul>\n<p><b>Prospek dan Rekomendasi<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Pengembangan kapasitas laboratorium genomik di institusi seperti LIPI dan BRIN perlu diperkuat. Kolaborasi internasional, misalnya dengan proyek <\/span><span style=\"font-weight: 400;\">Earth BioGenome<\/span><span style=\"font-weight: 400;\">, dapat memperkaya basis data genetik Indonesia (Margono et al., 2023). Di tingkat kebijakan, integrasi eDNA ke dalam Rencana Pembangunan Jangka Menengah (RPJMN) akan mendorong pendekatan berbasis sains dalam konservasi.<\/span><\/p>\n<p><b>Penutup<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Teknologi eDNA membuka era baru dalam biologi konservasi, menggabungkan presisi genetika dengan keberlanjutan ekologis. Untuk Indonesia, metode ini bukan hanya alat ilmiah, tetapi juga harapan dalam memitigasi krisis keanekaragaman hayati.
