400;\">Indonesia, sebagai negara tropis, merupakan hotspot penyakit bawaan nyamuk seperti demam berdarah dengue (DBD), malaria, dan Zika. Pada 2023, Kementerian Kesehatan RI melaporkan lebih dari 100.000 kasus DBD, dengan angka kematian mencapai 1.000 jiwa (Kemenkes RI, 2023). Upaya pengendalian konvensional seperti fogging dan penggunaan insektisida semakin terbentur dampak ekologis dan kemunculan resistensi pada nyamuk (<\/span><i><span style=\"font-weight: 400;\">Aedes aegypti<\/span><\/i><span style=\"font-weight: 400;\">)<\/span><span style=\"font-weight: 400;\">. Di sinilah teknologi <\/span>gene drive<span style=\"font-weight: 400;\"> yang berbasis rekayasa genetika muncul sebagai solusi revolusioner.<\/span><\/p>\n<p><b>Apa Itu Gene Drive?<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Gene drive adalah mekanisme yang memastikan gen tertentu diwariskan ke hampir 100% keturunan organisme, melampaui hukum Mendel (Esvelt et al., 2014). Teknologi ini memanfaatkan sistem CRISPR-Cas9 untuk menyisipkan gen yang diinginkan ke kromosom homolog, sehingga mengubah populasi target secara cepat. Pada nyamuk, gene drive dapat dirancang untuk menekan populasi (melalui sterilitas betina) atau membuatnya resisten terhadap patogen seperti virus dengue (Champer et al., 2021).<\/span><\/p>\n<p><b>Aplikasi Gene Drive dalam Pengendalian Nyamuk<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Uji laboratorium menunjukkan keberhasilan gene drive dalam mengurangi populasi nyamuk penular malaria <\/span><span style=\"font-weight: 400;\">(<i>Anopheles gambiae<\/i>) di Afrika hingga 99% dalam 12 generasi (Kyrou et al., 2018). Di Indonesia, riset serupa mulai diinisiasi oleh Lembaga Eijkman untuk memodifikasi nyamuk (<\/span><i><span style=\"font-weight: 400;\">Aedes aegypti<\/span><\/i><span style=\"font-weight: 400;\">)<\/span><span style=\"font-weight: 400;\"> agar membawa gen anti-dengue. Jika dilepas, nyamuk hasil rekayasa ini diharapkan memutus rantai penularan virus melalui populasi alami (Utarini et al., 2021).<\/span><\/p>\n<p><b>Tantangan dan Risiko<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Meski menjanjikan, gene drive tidak lepas dari kontroversi. Pertama, potensi <\/span>resistensi genetik<span style=\"font-weight: 400;\"> akibat mutasi acak pada target CRISPR dapat mengurangi efektivitasnya (Unckless et al., 2017). Kedua, dampak ekologis jangka panjang\u2014misalnya, terganggunya rantai makanan atau munculnya spesies invasif\u2014masih perlu dikaji. Ketiga, aspek etika: siapa yang berwenang memutuskan pelepasan organisme termodifikasi ke alam liar?<\/span><\/p>\n<p><b>Kesimpulan<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Gene drive menawarkan harapan besar untuk mengakhiri epidemi penyakit bawaan nyamuk di Indonesia. Namun, implementasinya memerlukan kolaborasi multidisiplin: ahli genetika, ekolog, dan regulator harus bekerja sama memetakan risiko, memantau dampak, dan menyusun kebijakan berbasis bukti.
