400;\">Keanekaragaman mikroorganisme merupakan indikator kunci kesehatan ekosistem (Hughes et al., 2001). Namun, pengukuran biodiversitas mikroorganisme secara kuantitatif memerlukan pendekatan yang menggabungkan teknik molekuler dan analisis ekologi. <\/span>PCR-RFLP<span style=\"font-weight: 400;\"> (<\/span><i><span style=\"font-weight: 400;\">Polymerase Chain Reaction-Restriction Fragment Length Polymorphism<\/span><\/i><span style=\"font-weight: 400;\">) menjadi salah satu metode andalan untuk memprofil komunitas mikroba, sementara <\/span>indeks Shannon (<i><span style=\"font-weight: 400;\">Shannon Diversity Index<\/span><\/i><span style=\"font-weight: 400;\">) memberikan kerangka matematis untuk mengkuantifikasi keragaman taksonomi. Artikel ini menjelaskan integrasi kedua pendekatan tersebut dalam studi biodiversitas mikroba.<\/span><\/p>\n<p><b>PCR-RFLP: Dari Amplifikasi DNA ke Profil Komunitas<\/b><\/p>\n<ol>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Amplifikasi Gen Target<\/b><span style=\"font-weight: 400;\">:<\/span><span style=\"font-weight: 400;\"><br \/>\n<\/span><span style=\"font-weight: 400;\">Gen penanda taksonomi (misalnya 16S rRNA pada bakteri) diamplifikasi menggunakan PCR dengan primer spesifik. Daerah hipervariabel seperti V3-V4 dipilih karena resolusi tingginya untuk membedakan spesies (Weisburg et al., 1991).<\/span><\/li>\n<li style=\"font-weight: 400;\" aria-level=\"1\"><b>Pemotongan dengan Enzim Restriksi<\/b><span style=\"font-weight: 400;\">:<\/span><span style=\"font-weight: 400;\"><br \/>\n<\/span><span style=\"font-weight: 400;\">Produk PCR dipotong oleh enzim restriksi (contoh: <\/span><span style=\"font-weight: 400;\">HaeIII<\/span><span style=\"font-weight: 400;\">, <\/span><span style=\"font-weight: 400;\">MspI<\/span><span style=\"font-weight: 400;\">) yang mengenali sekuens spesifik. Setiap pita (band) merepresentasikan OTU, dengan intensitas pita yang berkorelasi dengan kelimpahan relatif (meski perlu hati-hati karena bias PCR) (Schloss et al., 2009).<\/span><\/li>\n<\/ol>\n<p><b>Indeks Shannon: Kuantifikasi Keragaman Komunitas<\/b><b><br \/>\n<\/b><span style=\"font-weight: 400;\">Indeks Shannon (<\/span><span style=\"font-weight: 400;\">H\u2032<\/span><span style=\"font-weight: 400;\">) mengukur biodiversitas dengan mempertimbangkan <\/span>kekayaan spesies<span style=\"font-weight: 400;\"> (jumlah OTU) dan <\/span>keseragaman distribusi<span style=\"font-weight: 400;\"> (evenness). (2018) membandingkan komunitas bakteri tanah di area tercemar vs. alami menggunakan PCR-RFLP. (2020) mengaplikasikan PCR-RFLP pada gen 16S rRNA untuk menganalisis mikrobioma usus pasien lupus.
