neurodegeneratif yang menjadi penyebab paling umum dari demensia. Penyakit ini ditandai oleh akumulasi peptida amyloid beta (A\u03b2) di otak, terutama A\u03b240 dan A\u03b242, yang menjadi biomarker penting untuk diagnosis. Namun, deteksi dini penyakit ini sering kali terhambat oleh keterbatasan metode konvensional yang memerlukan waktu lama, biaya tinggi, dan sensitivitas yang rendah.<\/p>\n<p>Dalam upaya menghadirkan solusi yang lebih baik, Jia dkk. (2023) telah mengembangkan aptasensor berbasis CRISPR\u2014sebuah teknologi sederhana, cepat, dan terjangkau untuk mendeteksi biomarker A\u03b240 dan A\u03b242 secara kuantitatif dalam sampel cairan serebrospinal (CSF). Teknologi ini memanfaatkan spesifisitas tinggi aptamer terhadap biomarker A\u03b2 serta kemampuan CRISPR-Cas12a untuk menghasilkan sinyal fluoresensi saat mendeteksi target.<\/p>\n<h4><b>Keunggulan Aptasensor<\/b><\/h4>\n<p><span style=\"font-weight: 400;\">Aptasensor ini mampu mendeteksi A\u03b240 dan A\u03b242 dalam waktu kurang dari 60 menit dengan sensitivitas luar biasa, yaitu 1 pg\/mL untuk A\u03b240 dan 0,1 pg\/mL untuk A\u03b242. Sensitivitas ini jauh melampaui metode ELISA (Enzyme-Linked Immunosorbent Assay) yang lazim digunakan.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Untuk menguji keefektifannya, aptasensor ini telah diuji pada sampel klinis cairan serebrospinal. Hasilnya menunjukkan kemampuan untuk membedakan pasien AD dari individu sehat dengan tingkat akurasi tinggi. Kombinasi pengukuran konsentrasi A\u03b242 dan rasio c(A\u03b242)\/c(A\u03b240) memberikan klasifikasi diagnostik yang lebih presisi dibandingkan ELISA.<\/span><\/p>\n<h4><b>Implikasi dan Harapan<\/b><\/h4>\n<p><span style=\"font-weight: 400;\">Inovasi aptasensor berbasis CRISPR ini memiliki potensi besar dalam mendukung deteksi dini penyakit Alzheimer. Dengan waktu analisis yang singkat, sensitivitas tinggi, dan biaya rendah, teknologi ini berpeluang diadopsi secara luas di laboratorium diagnostik.
