400;\">Kulit merupakan salah satu material yang paling sering digunakan dalam berbagai artefak kuno. Identifikasi spesies kulit pada artefak ini memiliki signifikansi besar dalam memahami pola penggunaan dan persebaran material kulit di masa lalu. Namun, tidak adanya metode deteksi yang efektif menjadi tantangan besar dalam mempelajari\u00a0 penggunaan dan persebaran material kulit di masa lalu. Studi Huang dkk. (2024) menghadirkan pendekatan baru yang memanfaatkan kombinasi analisis proteomik-evolusioner dan teknik Enzyme-Linked Immunosorbent Assay (ELISA) untuk mengatasi kendala ini.<\/span><\/p>\n<h3><b>Metode dan Pendekatan Penelitian<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Penelitian yang dilakukan Huang dkk. (2024) diawali dengan analisis proteomik-evolusioner untuk mengidentifikasi komposisi protein dalam artefak kulit kuno. Dengan menggunakan basis data Swiss-Prot, sebanyak 154 protein berhasil diidentifikasi. Protein-protein ini kemudian dianalisis lebih lanjut melalui pendekatan evolusi molekuler, domain struktural, dan penjajaran sekuens (sequence alignment) guna memilih protein yang sesuai untuk proses lebih lanjut.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Dua protein, yaitu Kelch-like family member 17 (KLHL17) dan Nance-Horan Syndrome actin remodeling regulator (NHS), dipilih sebagai target untuk pembuatan antibodi. Afiliasi pengikatan antibodi terhadap protein ini diuji menggunakan pengukuran potensi antibodi dan teknik surface plasmon resonance (SPR).<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Setelah itu, dilakukan pengembangan dan optimalisasi metode ELISA untuk identifikasi spesies kulit kuno. Pengujian dilakukan untuk mengukur sensitivitas dan spesifisitas antibodi terhadap kulit dari berbagai spesies, termasuk kambing dan sapi.<\/span><\/p>\n<h3><b>Hasil Penelitian<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Metode ELISA yang dikembangkan menunjukkan performa yang sangat baik dalam identifikasi spesies kulit. Antibodi yang dihasilkan mampu secara spesifik mengidentifikasi kulit kambing dan sapi. Sensitivitas metode ini memungkinkan deteksi protein pada konsentrasi rendah, yang sering kali ditemukan pada artefak kuno.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Keunggulan utama dari metode ini adalah portabilitas, biaya yang relatif rendah, serta sensitivitas tinggi. Metode ini terbukti dapat diterapkan secara efektif pada identifikasi spesies kulit dalam konteks arkeologi.<\/span><\/p>\n<h3><b>Implikasi dan Kontribusi<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Penelitian ini tidak hanya menghadirkan metode baru untuk identifikasi spesies kulit kuno, tetapi juga memberikan wawasan penting dalam pengembangan metode berbasis protein untuk pelestarian dan analisis artefak budaya. Kombinasi pendekatan proteomik klasik dan teknologi ELISA memungkinkan penggalian informasi baru dari material kuno yang sebelumnya sulit diidentifikasi.<\/span><\/p>\n<h3><b>Kesimpulan<\/b><\/h3>\n<p><span style=\"font-weight: 400;\">Pendekatan berbasis proteomik dan ELISA yang diperkenalkan dalam penelitian ini menawarkan solusi efektif untuk mengatasi tantangan dalam identifikasi spesies kulit pada artefak kuno. Selain itu, metode ini berpotensi untuk diaplikasikan pada berbagai konteks lain dalam studi arkeologi dan konservasi artefak budaya.
