bioteknologi modern.<span style=\"font-weight: 400;\"> Seperti yang terlihat pada gambar pembuka, struktur kompleks protein antibodi ini telah menjadi kunci dalam berbagai terobosan bioteknologi. <\/span><\/p>\n<h3 class=\"fusion-responsive-typography-calculated\" style=\"text-align: justify; --fontsize: 36; line-height: 1.3;\" data-fontsize=\"36\" data-lineheight=\"46.8px\">Struktur Antibodi<\/h3>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Seperti yang terlihat pada gambar, antibodi memiliki struktur yang unik menyerupai huruf Y. Struktur ini terdiri dari dua rantai berat (heavy chain) dan dua rantai ringan (light chain). Ujung dari struktur Y ini disebut sebagai fragmen pengikat antigen (Fab), yang berfungsi untuk mengenali dan mengikat antigen secara spesifik. Bagian bawah dari struktur Y disebut sebagai fragmen kristalisasi (Fc), yang berperan dalam memicu respons imun lainnya, seperti aktivasi komplemen atau fagositosis.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Keunikan struktur antibodi terletak pada daerah yang disebut sebagai Complementary Determining Regions (CDR) atau daerah penentu komplemen. Daerah ini terletak pada ujung Fab dan memiliki variasi yang sangat tinggi, memungkinkan antibodi untuk mengenali bermacam-macam antigen yang berbeda. Fleksibilitas CDR inilah yang membuat antibodi menjadi molekul yang sangat spesifik dan efisien dalam menargetkan antigen.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Seperti yang telah kita bahas sebelumnya, antibodi memiliki struktur yang unik. Namun, tidak semua antibodi itu serupa. Berdasarkan asal dan sifatnya, antibodi dapat dibagi menjadi dua kategori utama, yaitu antibodi poliklonal dan antibodi monoklonal.<\/span><\/p>\n<h3 class=\"fusion-responsive-typography-calculated\" style=\"text-align: justify; --fontsize: 36; line-height: 1.3;\" data-fontsize=\"36\" data-lineheight=\"46.8px\">Perbedaan antara Antibodi Monoklonal dan Poliklonal<\/h3>\n<p style=\"text-align: justify;\">Antibodi poliklonal<span style=\"font-weight: 400;\"> dihasilkan oleh beberapa jenis sel B yang mengenali berbagai epitop (bagian spesifik dari antigen). Hal ini menyebabkan antibodi poliklonal memiliki beragam spesifisitas dan dapat berikatan dengan berbagai bagian dari suatu antigen. Meskipun memiliki afinitas yang lebih rendah dibandingkan antibodi monoklonal, antibodi poliklonal sering digunakan dalam berbagai aplikasi, seperti Western blot dan imunohistokimia.<\/span><\/p>\n<p style=\"text-align: justify;\">Antibodi monoklonal<span style=\"font-weight: 400;\">, di sisi lain, dihasilkan oleh satu jenis sel B yang hanya mengenali satu epitop spesifik pada antigen. Hal ini membuat antibodi monoklonal memiliki spesifisitas yang tinggi dan afinitas yang kuat terhadap antigen target. Karena sifatnya yang homogen, antibodi monoklonal banyak digunakan dalam terapi kanker, diagnosis penyakit, dan penelitian dasar.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Proses produksi antibodi poliklonal dan monoklonal juga berbeda. Antibodi poliklonal umumnya diperoleh dengan imunisasi hewan (seperti kelinci atau kambing) dengan antigen target. Serum hewan yang mengandung campuran antibodi poliklonal kemudian dapat diisolasi dan digunakan untuk berbagai aplikasi.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Sebaliknya, produksi antibodi monoklonal melibatkan teknik hibridoma, di mana sel B yang menghasilkan antibodi spesifik difusi dengan sel myeloma (sel kanker) untuk menghasilkan hibridoma. Hibridoma ini kemudian dapat diklon untuk menghasilkan sejumlah besar antibodi monoklonal yang identik.<\/span><\/p>\n<h3 class=\"fusion-responsive-typography-calculated\" style=\"text-align: justify; --fontsize: 36; line-height: 1.3;\" data-fontsize=\"36\" data-lineheight=\"46.8px\">Domain Penggunaan Antibodi<\/h3>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Antibodi umumnya digunakan dalam tiga bidang: riset, diagnostik, dan terapeutik. Salah satu contoh penggunaan antibodi di bidang riset adalah penggunaan antibodi dalam teknik <\/span>ELISA (Enzyme-Linked Immunosorbent Assay)<span style=\"font-weight: 400;\">. ELISA memungkinkan kita untuk mengukur jumlah suatu antigen secara kuantitatif dalam sampel biologis. Antibodi spesifik yang melapisi permukaan pelat ELISA akan berikatan dengan antigen target, dan deteksi kompleks antigen-antibodi ini dapat dilakukan secara enzimatis.<\/span><\/p>\n<p style=\"text-align: left;\"><span style=\"font-weight: 400;\">Selain ELISA, antibodi juga digunakan dalam teknik <\/span>Western blot<span style=\"font-weight: 400;\"> untuk memisahkan dan mengidentifikasi protein spesifik dalam sampel. Antibodi yang spesifik terhadap protein target akan berikatan dengan protein tersebut setelah proses elektroforesis dan transfer ke membran. Teknik lain yang melibatkan penggunaan antibodi adalah <\/span>imunohistokimia<span style=\"font-weight: 400;\">. Teknik ini memungkinkan kita untuk memvisualisasi lokasi protein spesifik dalam jaringan menggunakan mikroskop. Antibodi yang berlabel dengan molekul fluorescent atau enzim akan berikatan dengan protein target di dalam jaringan, sehingga lokasi protein tersebut dapat diamati.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Dalam bidang diagnostik, antibodi juga berperan dalam berbagai jenis metode deteksi penyakit. <\/span>Tes cepat (<i>rapid test<\/i>)<span style=\"font-weight: 400;\"> atau <\/span><i>lateral flow assay<\/i><span style=\"font-weight: 400;\"> adalah salah satu contoh aplikasi antibodi dalam diagnosis. Tes ini sering digunakan untuk mendeteksi antigen virus atau bakteri secara cepat dan mudah. Prinsip kerjanya mirip dengan ELISA, namun dalam format yang lebih sederhana. Antibodi yang terimobilisasi pada strip tes akan berikatan dengan antigen target dalam sampel, menghasilkan garis berwarna yang mengindikasikan hasil positif.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Selain tes cepat, antibodi juga digunakan dalam <\/span>ELISA (IVD)<span style=\"font-weight: 400;\"> untuk mendiagnosis berbagai penyakit, seperti penyakit infeksi, penyakit autoimun, dan kanker. Dengan menggunakan panel antibodi yang spesifik, kita dapat mendeteksi keberadaan autoantibodi atau biomarker penyakit lainnya dalam sampel pasien.<\/span><\/p>\n<p style=\"text-align: justify;\"><span style=\"font-weight: 400;\">Salah satu perkembangan paling signifikan dalam bidang bioteknologi adalah penggunaan antibodi sebagai terapi. <\/span>Antibodi monoklonal<span style=\"font-weight: 400;\"> telah menjadi terapi pilihan untuk berbagai jenis kanker. Antibodi monoklonal yang dirancang secara khusus dapat menargetkan sel kanker secara spesifik dan memicu kematian sel kanker atau menghambat pertumbuhan tumor. Contoh obat yang mengandung antibodi monoklonal adalah <\/span>adalimumab<span style=\"font-weight: 400;\">.
